Can EPO server 2.5.1 update DATs on 4.5.1 and 7.1.0 clients simultaneously?

I would've thought so, mate.

I've never tried it though, so I couldn't actually say for sure.

It certainly can. VirusScan 7 requires extra files to update though.

use ftp.nai.com/common updater for the DAT updates.
