Question: I want to setup a folder called support (and others eventually) so that a visitor can browse to

http://support.mydomain.com

What's the best method of doing this?
Do I need a seperate IP for each?

create a site with a host header for the sub-domain then add the sub-domain to your DNS server or make sure that the DNS server supports wildcards. (*.domain.com)
