I have a computer that has an addition to the taskbar called search assistant. I've ran AV and Spybot and can't seem to figure out how to get rid of it. What do I have to do to get rid of it.
It worked fine for my pesty, unwanted search toolbar...

There are different 'search assistants'. If it is the 180 version you will likely need to run a minimum of Adaware and maybe a second opinion from Spybot. These antispyware tools remove the related junkware from your computer. The latter two (2) also have free 'audit' tools which would enable you to see if any 'search assistant' is still in evidence:

I would modify those instructions to run cwshredder, spybot and adaware in safe mode and reboot after each one. Working in safe mode gives the minimum of other stuff running and has improved these applications success in cleaning up some of the more tenacious items.
