If I send an E-mail the reply-to Adress is my internal e-mail-adress.

For example: Exchange has the domain "domain.com" but my email-adress is office@t-online.de
my Secretary has secretary@t-online.de
but Exchange uses as reply-to secretary@domain.com

Where can I tell Exchange which reply-to adress it want to use.

In Active Directory, go to the properties of the account for your secretary. Go to the email properties tab, highlight the email address you want to be the reply-to, and click the "Set as Primary" button.
