Outbound mail inexplicably stops working!

Here's the 911:
Several times since we have installed Exchange2000 Server, the outbound mail has stopped working. Users will get the error message that their mail did not reach some or all of the intended recipients or that it was being delayed and will be retried. I've forced the connection but that doesn't work. When I reboot the Exchange Server, the outbound mail delivery works fine again until the next time it happens. Also, the outbound mail has to run through ISA server first (on another machine) before going out. Anyone aware of or have any solutions to this problem?
 
ISA is on a different box right?

Do you point to Windows 2k DNS only on the server if not change that. Dan

Yes, ISA is on a different box. Also, DNS is also running on the ISA server. However, if it can't resolve any DNS queries, it goes out to the internet using forwarders. Any more suggestions?
