Does local policy on DC affect domain user accounts?

We've setup our domain policy with no password expiration, however some users are saying that they still get prompted to change their password. When I run RSOP, the results say not configured, which is correct. But when I look a the local policy on the DC (there's only one in the domain) it's configured to maximum age of 42 days. Surely this wouldn't affect the accounts that exist on the domain? If so, then what would happen if I had two DC's and the local policies were different?
