Start with a check on the queues to see WHY it cannot send.
Also check the Eventlogs to see if anything shows.

check if port 25 is open by attempting a loop-back telnet on port 25.
What antivirus scanner are you using on the machine? We had an issue with McAfee Virus Scan 8.0i closing port 25 automatically.
Port 25 is incoming to receive. What are you all refering too?
It can be closed, and you can still SEND then!
