Blocking Spam

We seem to be getting quite a lot of spam even though we have a firewall, webshield etc in place. What I would now like to block, is spam coming in that is making use of our domain name. ie. Call us test.com. I would like to block any messages from test.com accounts that are NOT registered on our system- Not valid users. Is this possible?

I know you can block the entire domain name from coming in, but we will still require registered users being able to send from outside, in.
