On my Inventory Negative Tier report an item has Quantity on Hand 0 (zero) but the Tier Quantity for the same item shows -2 (negative 2)
Why does this happen and how can I correct it?

Just run the Negative Tier Adjustment. This happens because you may have sold an item when you had zero on hand resulting in a negative tier which you see. Later you either received in or adjusted in the item. The next result of the positive tier is a Quantity On Hand of Zero (0). What the negative tier adjustment does is apply the negative tier to the positive tier so everything is zero and they are purged at month end.
