Website stops working when I change access right of root folder

Hi friends,
Iam facing some problems while changing access rights of the root directory of our website.
Due to security reasons we want to give more rights on the root directory.The moment I give more access rights to the root directory,the website itself stop working( it fails to display the pages)
Can anybody suggest some solution ?

I don't see how granting access could cause a problem, unless you removed the evryone group and didn't give access to the IUSR_machinename default IIS user. Or maybe your using NT Authentication, at which point you would need the everyone group to have have read, maybe read and execute, etc. You also didn't mention if you were setting the permissions from explorer (rt clicking on folder) or if you were doing it from the ISM in admin tools.
