I have a single domain environment with 1 PDC, 1 BDC, and 1 File Server. I have 200 Employees in a central location and 100 employees at 18 remote locations. I would like for the 200 centrally located users to authenticate to the PDC and the 100 remote users to authenticate to the BDC. All employees will share the single file server. How can I specify which server for the individual employees to authenticate to?

Probably not a good idea. What happens when one of these DC servers are taken off-line or fail? You'll have a lot of people complaining that they can't log on. Why would you want to do this?
