Select unmatched records using a query

Does anyone know if it's possible for a query to show only the unmatched rows from a table?

Example: If table one has the field [Letter] and there are 5 records, A-E, and table two has a similar field with the 3 records A-C I would like the query to show only the records "D" & "E".

Normally I would do this in excel, matching records and filtering for where no match is returned. The amount of data I am using makes excel hang, so I've had to turn to access.

 
Will have to do a bit of in-depth checking, but the number of records shown is definitely in the right ballpark for what I was expecting. Thanks a mil - that will save me a LOT of time.
