Does it matter which of the two SATA ports that you plug your hardrive into? My motherboard has two one is marked 0 and the other 1.I am only installing one drive,no raid..

No it doesn't matter but you would normally start with the lowest number first and in your case this is SATA 0.
Be sure to install your SATA controller drivers which you should find on your motherboards driver disk or downloadable from the motherboard manufacturers website.
Initialize and format in disk management using tools.
