This may not be the place to ask, but will a roaming profile work on both a WinXP machine and a Win2k machine?

We use roaming profiles and Win2k / XP clients without any problems. There we're problems with NT/2000 as they store the profiles in a different local folder, but with 2000/XP i haven't had any problems.

do you use outlook on those machines as well? We want to use Outlook 2003 with Windows 2000 or XP in a 2k3 environment

Only a couple of users use Outlook 2000 and it seems to work just fine, put the PST file on the server and make sure it doesn't roam as it can get big. (You don't want it to download at every logon)

can this be globally set somewhere, like when i create the first profile? Or do i have to fix each profile?

I don't use Exchange but as far as i know that all depends on how you set it up. If you use it as a pop3 server then yes (i imagine most don't) but using Outlook the mail can be stored in the Exchange server store.
I would ask that in the Exchange server forum, i'm pretty sure you can roll out Exchange settings to multiple users.
