Can someone tell me what this message means when there is paper in the both trays? How can i fix this?

"Cleaning sheet needed - Load letter size paper"

What printer do you have? Typically this means that the upper tray on say a 840/850/860 is defaulted for the cleaning sheet, which needs to be letter size. Check the tray size sensing plugs on the right side of the tray, I am guessing the top one is missing that is why it is asking for the letter size media to be loaded.
