PNG transparency not transparent

I know I must be missing a step when optimizing in ImageReady, but my PNG files are not transparent when brought into either an image placeholder or a floating box. I am checking 'transparent' and 'none' for matte when optimizing. I need to use the PNG format, as the image has a multilevel transparency (feathered area) and I'm placing it over another image. Sort of a movie screen effect for remote rollovers. These are fairly small images, less than 50px x 50px. They look transparent when they are placed and positioned, but in preview (and browser) the box is gray around the image. What am I missing? Thanks in advance.
