/var should be pretty large to accomodate Web and Mail if you stick to the default locations. Depending on size and volume of both you should accomodate for more space than the default provided by tools like Disk Druid. 10Gb should do it in most small business cases.
The web stuff by default lives in /var/www.
Mail boxes live in /var/spool/mail, sendmail spools to /var/spool/mqueue.
If you are going to use maildirs instead of mailbox then /home should be sufficiently large enough to accomodate mail for all the users.
The best way to go about it is to rather allocate most of your disk to a Logical Volume Manager (LVM), then create file systems in the LVM. The trick is to not allocate all of the LVM space to the file systems, if in the future you require to increase a FS it's a pretty easy an painless task to increase a FS using the free LVM space.
