Microphone doesn't work

I am trying to connect my microphone to my computer but what happens is that I can hear the person on the other side perfectly but she can't. Always when I open the right icon below the screen my volume is mute and I change and as soon I come back to verify turns back to mute. I open the details also but nothing happens. I also went to a store and was informed that I bought a very good microphone and the guy placed into one of the computers store and it worked perfectly. Do you have any idea what can be the problem?? I read the help and follow all the instructions but still nothing. So my last hope was maybe you.

Sounds like hardware, is your sound card compatable?

Actually what I can tell you is that I love computers but I hate them when I have to solve a problem because I don't understand anything about it. Sorry the ignorance, but what is sound card? How do I know if it's compatable?
