You sure can, by using the 'show-hide layers' behavior.
Start by naming your layers (it makes life easier), then using the show-hide behavior (F8 will open this pallet)set the layer to hide 'onLoad'. then with your button selected use the same behavior to 'show' the layer on mouseover or mousedown whatever you want.
