Hi,
If you have merges all your layers and there is only one layer present on your layers palette, then the answere to your question is no. Once merged, saved and closed, you can't undo it.

Spyderix is quite right to say once "saved and closed" that the end of your layers.
Just a note of clarification, if you flatten an image, you can recover prior to saving and closing your file by "undoing the flatten" or by dragging the flatten from history to the trash can.
