We have a few machines at work with an annoying issue.

When the computers go into standby, the only way to wake them is by pressing the power button on the computer.

I've enabled keyboard and mouse wakeup in the BIOS. Motherboard is an Asus P5KPL-SE with the most recent BIOS (0506).

When you tell Windows to sleep, it powers off the mouse and the keyboard, and because of this it's not possible to wake the machine. Is there a setting in Windows that's doing this or is it possibly a BIOS issue?

Are there power settings for the Keyboard, and Mouse, via their respective Power Options in their Properties in the Device Manager?

The only power setting is Allow this device to bring the computer out of standby and it's checked on both devices.

Have a look at Powercfg /? from a Command Prompt. There are settings in there for things like -DEVICEENABLEWAKE,
-LASTWAKE, and other interesting bits and pieces.

Do these machines come with any installed OEM Power Control software?

There's no OEM control software. I did try changing some settings with dumppo to no avail.

I did notice that most of the affected workstations seems to be the same motherboard with this issue. I'm starting to lean towards a BIOS bug.

I was about to make the same suggestion - BIOS issues.
