Is it possible top block lookups of a particular site?

I think this is a dns problem, but I can't figure out how to figure it out. There is a particular site which is inaccessable from our private network. The network is firewalled, but does not block either udp or tcp ports53. The firewall and screening routers do block particular addresses, but, they don't block lookups. I usually test for a blocked site by going to command prompt and ping the blocked site. The address is usually returned by a dns lookkup, but not the ping.

However, for the site no address is returned. My question is: How is it possible to block the lookup of only a single site without affecting the lookups of other sites?
 
By making a hosts entry for the site and referencing your localhost address.
 
Wouldn't that return the address of localhost 127.0.0.1? I'm receiving no return address.
