Having created a single domain with several domain controllers using sites, is it possible to lock down the sites - ie a user at one location can only logon to the site appropriate to them and get access to files/data only from their own domain controller. If possible, how is this done?

I dont believe that restricting users based on site is possible. You could go into each user account and only give them access to log onto a list of computers at their site and set NTFS permissions on the files to only allow the users of each site to access the data of that site.

What you probably really want to do is set up multiple domains (one for each current site) to be part of the same forrest. You could then set restriction on each domain.
