Put this in the Target box. The C:\ can be replaced to the place where you want to open it.

I do not think you can change the Winkey+E defaults.
You can create a shortcut, and use that instead following the advice above.
right-click the Desktop, New, Shortcut, (to run the shortcut wizard) and set the target as explained above. But this does nothing for Winkey+E
