EPO 3 and ISA server W2K

Not recommended. ISA Server has too many connections/possibilities to get infected potentially and you probably DO NOT WANT ePO to be the source of infections 
Suggest a separate server for ePO, and if you can spare it, a separate SQL Server connected directly to the ePO server via cross-cable. This will ensure that the connections between the ePO server and the database is always on and quick.
