Go to My Computer - Manage. Look in Shared Folders - Open Files. See if the file is listed in there. If it is, right-click and close it. See if you can delete it then.
Also try logging in as administrator and taking ownership of the folder in Security - Advanced.
We have also recently had something like this problem. A client could connect to the Citrix Server/TS once, then when trying to connect again would get similar error messages or a Dial-Up connection error (even though it's through a LAN :O))
I had to give Full Control to the security...
Definately a print spooler problem, especially if you're running Win2K SP3. I put a scheduled job on each of our servers to restart the print spooler service every hour when we started getting this problem. Seems to have "worked"...but I think microsoft has released a hotfix for this...
As soon as I get back to work tomorrow I'll check this out for you...I spent a couple of days last week sorting this out our farm. I'd tell you now...except it's been a good weekend and I can't remember half of what I did! :O)

Basically if your server is running fine on SP2 then don't upgrade.
There is currently a fourteen page list of post-sp3 hotfixes. If you don't need to upgrade...don't.
I had sort of this problem...had COM port mapping working and transmitting data then all of a sudden...bang, it stopped. Not sure what caused it but made it so the user could get to a cmd prompt, did 'net use' and I could see the COM port was actually mapping but not allowing data to be passed...
