What is the easiest way I can set up a large number of user's home folders (including security settings) both on the file server and on their account properties in User Manager?

Tell me there is a easy way??

Use the %username% variable in the path for their folder mapping in user manager. e.g \\servername\sharename\%username%

For new users this will automatically create and set the security so no-one else can access them.

If your users accounts already exist then you will need to use a script to set the security on their folders.
Not sure if something like dameware or hyena could do this for you

Thanks SjrH - I actually tried this but it errorerd and said it could not create the folder and that I have to create it manually - ever seen that before?

You usually only get that error if either the folder doesn't exist, isnt shared (as per the \sharename), or you have already created the folder manually.

Aaah - The PARENT folder has to be shared - silly but true
